Just the chemical bonds and intermolecular forces, combined with moving the bits around following a formula. However, you could turn fat into a hydrogen-propelled diamond bullet.
Absolutely, and it is stated in FMA that that activation energy is theoretically derived from tectonic potential energy, somehow (although, of course, that turned out to be a convenient lie, in-canon)

Bonds take energy to break and make. Some is released when they break. And those forces take energy to overcome.
